I just took a look at mine. The top goes to the manifold, bottom to the water pump.

According to the 67-72 shop manuals, the hose coming from the intake (8 cyl) runs into the lower heater core fitting, the upper heater core fitting runs into the water pump (non a/c) or to the radiator (with a/c).

I have a 70 & 72 sitting out front.
No matter which one I looked at the bigger of the two hoses goes to the water pump.

Does it matter if it is backwards? My guess is yes. The heater core might never be 100% full if you try to run it backwards/upside down. If the 3/4" hose is at the bottom it will be able to empty the core faster than the 5/8" hose can fill it. That might also mean listening to the sound of running water as it gurgles on through the heater core. On the cold days of MN I would think it wouldn't put out the heat like it should either.
